First, we assessed spatial alignment using more biologically detailed microstructural metrics; second, we compared age-related patterns between markers, anticipating a strong relationship between measures primarily influenced by similar myelo- and microstructural modifications. The CIVET 21.0 pipeline facilitated the generation of cortical surfaces from MRI images of 127 healthy individuals, aged 18 to 81, used in the derivation of cortical MRI markers. Their gross spatial patterns were analyzed alongside cell-type densities derived from gene expression, histology-based cytoarchitectonics, and the quantitative R1 maps collected from a segment of the participants. We then compared age-related shifts in the morphology, directionality, and spatial spread of the linear age effect for the markers. The gross anatomical distribution of cortical MRI markers exhibited a correlation, in general, more strongly to myelin and glial cells than to neuronal indicators. Results from comparing MRI markers demonstrated a notable consistency in spatial distribution across groups, but showed mostly different age trajectories for the shape, direction, and spatial distribution of the linear age effect. We observe that the microstructural factors driving the spatial distribution of MRI cortical markers may exhibit differences from the microstructural modifications associated with aging and impacting these markers.

HRAS pathogenic variants, activating postzygotically, were previously found in nevus sebaceous (NS), keratinocytic epidermal nevus (KEN), and diverse enteric nervous system (ENS) conditions like Schimmelpenning-Feuerstein-Mims and cutaneous-skeletal-hypophosphatasia syndrome (CSHS). HRAS-related enteric nervous system (ENS) conditions, characterized by KEN, can display a range of skeletal manifestations, from localized bone dysplasia to the more extensive fractures and limb deformities frequently seen in CSHS. We initially link HRAS-related ENS with auricular atresia, thereby broadening the disease spectrum to include first branchial arch defects when the mosaic variant is present. This report illustrates, for the first time, the simultaneous occurrence of verrucous EN, NS, and nevus comedonicus (NC), suggesting the likelihood of mosaic HRAS variation as an underlying factor in NC.
